Abstract

Objective To investigate the effect of stellate ganglion block (SGB) on endothelin-1 (ET-1) and calcito-nin gene-related peptide (CGRP) in the plasma and hippocampus of rat in cardiopulmonary bypass (CPB).Methods Thirty male Sprague-Dawley rats were randomly divided into three groups (n=10):the sham operation group (group S), group CPB and group SGB.CPB models were established in the group CPB and SGB, and rats in the SGB group received SGB before CPB.Venous blood samples were taken at 0 h ( T0 ) , 1 h ( T1 ) and 2 h ( T2 ) after CPB for determining the ex-pression of ET-1 and CGRP in plasma by ELISA.And the hippocampus was isolated for determining the expression of ET-1 and CGRP by ELISA.The pathological change of pyramid cells in the CA1 region of hippocampus was also examined. Results Compared with group S, the expression of ET-1 in both plasma and hippocampus was significantly increased at T1 and T2 in the groups CPB and SGB (all P<0.05), and the expression of CGRP in plasma was decreased in the groups CPB and SGB (all P<0.05), while in the group SGB, the expression of CGRP in hippocampus was significantly in-creased.HE staining showed that there were different degrees of damage of hippocampus nerve cells in the groups CPB and SGB.Compared with group CPB, the expression of ET-1 in both plasma and hippocampus was significantly decreased at T1 and T2 in the group SGB (all P<0.05), while the expression of CGRP was significantly increased in the group SGB (all P<0.05), and HE staining showed that the damage of hippocampus nerve cells in the group SGB was less than that of the group CPB.Conclusion SGB can improve the brain tissue perfusion and reduce cerebral damage of rat in CPB by regula-ting the expression of ET-1 and CGRP in the plasma and hippocampus.
