Anti-tumor effect of calculus bovis cultivated by glucuronidase on mice sarcoma S180 cells

Abstract
Objective To investigate the anti-tumor effect of calculus bovis cultivated by glucuronidase ( CBCG ) on routine sarcoma S180 cells in mice and the related molecular mechanism.Methods Fifty Kunming mice were selected to es-tablish the S180 tumor-bearing mouse models, and then they were randomly divided into 5 groups, 10 in each:the high-dose, medium-dose and low-dose CBCG groups (which were respectably administered with 2 g/kg, 1g/kg and 0.5 g/kg), natural calculus bovis (NCB) group (which was administered with 1 g/kg NCB) and the model group (which was given 0.1 g/L starch solution).The mice were sacrificed after treatment with drugs for 10 days.The tumor tissues were corrected to calcu-late the tumor inhibition rate in each group except the model group.Western blotting was used to detect PTEN, AKT, P-AKT (Thr308)/AKT, P-AKT (Ser473)/AKT in PIEK/AKT pathway and the apoptosis-related protein expression of Bax and Bcl-2 in each group.Results CBCG inhibited the tumor growth of tumor-bearing mice with a dose-dependent manner ( all P<0.05).No significant difference was found in the inhibitory rate between the NCB group and the high-dose CBCG group. Compared with the model group, the expression of PTEN and Bax was increased but the expression of P-AKT (Thr308), P-AKT (Ser473) and BCL-2 was decreased in the high-dose, medium-dose and low-dose CBCG groups.Conclusion CBCG inhibits S180 growth of tumor-bearing mice, and the possible molecular mechanism may be related to the PI3K/AKT pathway.关键词
