The effects of Fn14 on metastasis in human epithelial ovarian cancer cells

Abstract
Objective:To investigate the role of fibroblast growth factor-inducible 14 (Fn14) in metastasis of human epithelial ovarian cancer cells. Methods:Human epithelial o-varian cancer cells HO8910 and high-metastatic human epithelial ovarian cancer cells HO8910-PM were collected. Fn14 protein expression levels in HO8910,HO8910-PM and HO8910-PM transfected with Fn14 plasmid were detected using Western blot. The expressions of Slug and MMP-9 were determined by Western blot. The migration and invasion abilities of cancer cells were assessed using transwell assay. Results:Both migration and invasion numbers of HO8910-PM cells were higher than that of HO8910 cells (P<0. 05). Compared with HO8910 cells,the expression of Fn14 in HO8910-PM was decreased obviously ( P<0 . 05 ) . Fn14 protein level was significantly increased in HO8910-PM cells when Fn14 gene was transferred. Fn14 gene transfer could decrease the ability of migration and invasion,as well as protein expression of Slug and MMP-9 ( P<0 . 05 ) . Conclusions:Fn14 could inhibit the metastasis of the epithelial ovarian cancer cells through regulating Slug and MMP-9 expression,suggesting that Fn14 might be a no-vel target for the therapy of human epithelial ovarian carcinoma.关键词
