Abstract
Objective To investigate the protective effect of paeoniflorin on acute liver injury in mice.Methods The mice were randomly divi-ded into 5 groups:control group, model group, paeoniflorin (Pae) alone group, as well as groups of paeoniflorin and lipopolysaccharide ( LPS).Animals were injected LPS once daily for 7 d to establish the LPS infla-mmation model in mice except the control and paeoniflorin group.The activation of tumor necrosis factor ( TNF -α) and interleukin -1β( IL-1β) in serum was evaluated by Enzyme -linked immunosorbent assays (ELISA).Then the alanine aminotransferase (ALT), aspartate aminotransferase ( AST ) , malondialdehyde ( MDA ) , superoxide dis-mutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione (GSH), nitricoxide (NO) and inducible nitricoxide synthase ( iNOS) were detected by the colorimetric method.The levels of nuclear factor-κB ( NF-κB) , IκB-α, p-IκB-αprotein in subcellular frac-tions of hepatic tissue were detected by Western blot .Results Compared with model group , the levels of TNF -α, IL-1β, ALT, AST, MDA and NO, and the activities of iNOS reduced in 30 mg· kg -1 Pae group;while the activities of antioxidant enzyme ( SOD, CAT and GSH -PX) and the level of GSH significantly increased in 30 mg · kg -1 Pae group. Furthermore, Compared with model group , the degradation of IκB-αdecreased and the activation of NF -κB was in-hibited in 30 mg· kg -1 Pae group.Conclusion The paeoniflorin has the protective effects on acute liver injury.关键词
