Deficiency of MTMR14 Promoted the Proliferation and Differentiation of Mouse Myoblasts

Abstract
To investigate the role of myotubularin related protein 14 (MTMR14), a novel phosphoinositide phosphatase, in regulating myogenesis, the effects of MTMR14 depletion on the structure, myoblast differentiation and proliferation were evaluated using tissue biopsy and cell culture techniques. The results demonstrated that the structures of both gastrocnemius and soleus muscles in MTMR14-depleted mice were different from their wild type ( WT) littermates. Compared to the WT control, the myoblast of MTMR14-depleted mice showed accelerated proliferation and differentiation into myotubes in culture. The depletion of MTMR14 also induced an increase of the number of nuclei in myotubes. In conclusion, the deficiency/depletion of MTMR14 led to the abnormal structure of skeletal muscles, and disrupted the proliferation and differentiation of myoblasts during skeletal myogenesis.关键词
