Abstract
Objective:To study the clinical significance of high performance liquid chromatography (HPLC) in the determination of puerarin content in Puerarin Injection.Methods:The puerarin content in Puerarin Injection was determined by Shimadzu Prominence LC-20AT HPLC.Merk,Purospher STAR RP-C18 (250×4.6nm,5μm) chromatographic column,1％ of acetic acid aqueous solution and methanol were matched as mobile phase according to the ratio 25:7.The column temperature was 29degrees,the flow rate was1.0ml · min-1,the sample size was10μl and detection wavelength was at 250nm.The value of HPLC in the determination of puerarin content in Puerarin Injection was analyzed.The differences in puerarin content of Puerarin Chloride Sodium Injection,Puerarin Glucose Injection and Puerarin Injection made by different manufacturers were observed.Results:The precision,stability and reproducibility of HPLC in detection of Puerarin Injection were better.The average recovery rate was 98.76％ and RSD was 1.3％.There was certain difference in puerarin content of Puerarin Injection made by different manufacturers,which were 112.36mg.ml-1,111.42mg.ml-1 and 113.58mg.ml-1 respectively,but the overall difference was small.However,the differences in puerarin content of Puerarin Glucose Injection,Puerarin Chloride Sodium Injection and Puerarin Injection were relatively larger,but all met the specification standard.Conclusion:HPLC in the determination of puerarin content of Puerarin Injection is simple,rapid and accurate.It can be used as the effective way to detect the puerarin content of Puerarin Chloride Sodium Injection,Puerarin Glucose Injection and Puerarin Injection.And there are differences in puerarin content of different batches of Puerarin Injection,but the differences are small.The differences in the puerarin content of Puerarin Chloride Sodium Injection,Puerarin Glucose Injection and Puerarin Injection are larger,which needs the clinical attention.关键词
