GA Biosynthetic Enzyme GA3OX1 Required for Shade Avoidance in Arabidopsis

Abstract
Shade avoidance response is a plastic response induced in the shade-avoiding species in response to shade.It is an important adaptive response that allows plants to compete for light when it is under or near vegetative shade.In this study,we isolated a sav8 (shadeavoidance8)mutant that was defective in multiple shade avoidance responses.We characterized the phenotypes of sav8 and conducted map-based cloning to identify the mutation in it.Compared with wild type (Col-0),sav8 had shorter hypocotyls under sim-ulated shade and dark,and adult sav8 was smaller,more branchy,and with reduced apical dominance.Under simulated shade,sav8 was hypersensitive to gibberellins (GA).Through map-based cloning,we identified that sav8 contains a C-to-T mutation in a GA synthetic gene GA3OX1 (AT1g15550),which converted Gln309 to a stop codon and encoded a truncated protein.TAA1 (tryptophan aminotransferase of Arabidopsis 1 )is a key gene required for the shade avoidance responses.Our study showed that GA3OX1 was up-regulated by shade in a TAA1-independent manner.Taking together,our study demonstrated that GA3OX1 is a key gene required for shade avoidance response.关键词
