补肺益肾方对幼龄哮喘豚鼠模型IL-5信号通路的调控机制
Regulating mechanism of traditional Chinese medicine of invigorating lung and nourishing kidney on IL-5 signal pathway of young guinea pigs with asthma

Abstract
Objective To study the effect of the traditional Chinese medicine of invigorating lung and nourishing kidney on interleukin-5 level and expression of IL-5 receptorαmRNA in eosinophils( EOS)of bronchoalveolar lavage fluid( BALF)of young guinea pigs with asthma and to explore the mechanism of the traditional Chinese medicine of invigorating lung and nourishing kidney in promoting eosinophil apoptosis by IL-5 signal pathway. Methods The 24 young guinea pigs were divided into three groups randomly,normal control group,model group and the traditional Chi-nese medicine group. Asthma model of guinea pigs were sensitized by ovalbumin. EOS was purified from BALF. The content of IL-5 in BALF was detected by enzyme-linked immunosorbent assay( ELISA ). Apoptosis was detected by TdT-mediated dUTP nick end labeling(TUNEL). Expressions of IL-5 IL-5Rα mRNA were measured by bridization. Results The content of IL-5 in model group was higher than that of normal group( P﹤0 . 01 ),while the content of IL-5 in the traditional Chinese medicine group was lower than that of model group( P﹤0 . 05 ). EOS apoptosis in model group guinea pig were significantly decreased,while EOS apoptosis in the traditional Chinese medicine group guinea pig were higher than that of model group( P﹤0 . 01 ). Expressions of IL-5 RαmRNA in EOS from model group were re-markably lower than that of normal group( P﹤0 . 01 ),while expressions of IL-5 RαmRNA in EOS from the tradition-al Chinese medicine group were increased( P﹤0 . 05 ). Conclusion The traditional Chinese medicine of invigorating lung and nourishing kidney promotes eosinophil apoptosis by decreasing the content of IL-5 in airway and increasing expressions of IL-5 R α mRNA. So it can relieve airway inflammation and prevent asthma.关键词
