Abstract
Objective To investigate the influence of galectin-3 (Gal-3) on biological behaviors of epithelial ovarian cancer ( EOC) cells.Methods After siRNA-mediated knockdown of Gal-3 and plasmid transfection-media-ted overexpression of Gal-3 in OVCAR3 cell line, CCK-8 kits were used to measure proliferation in OVCAR3 cells. Wound healing test and transwell were used to observe cell migration and invasion ability.Western blot was used to detect the protein expression levels of c-myc, cyclin D1, matrix metalloproteinase-7 (MMP-7).The control group was trea-ted with negative control siRNA at 50 nmol/L.Results The capacity of proliferation, migration, invasion of EOC cells with down-regulated Gal-3 were decreased compared with the control group (P<0.001).The protein expression levels of c-myc, cyclin D1 and MMP-7 were decreased;Overexpression of Gal-3 increased cell proliferation,migration and invasion,the differences among them and control group were statistically significant (P<0.05),the protein expression lev-els of c-myc, cyclin D1 and MMP-7 were increased.Conclusions Gal-3 promotes proliferation, migration and inva-sion capability of EOC cells.Gal-3 may be a promising target of EOC targeting therapy.关键词
