摘要

Abstract

Objective To examine the effect of miR-143 on the proliferation of acute leukemia cells HL-60 by targeting KRAS.Methods Luciferase reporter gene assay was used to detect the tar-geting role of miR-143 to KRAS in HL-60 cells.RT-PCR was used to detect the mRNA expression of miR-143 and KRAS in HL-60 cells.Western blotting was employed to detect the protein expression of KRAS, and MTT method to detect the proliferation of HL-60 cells.Results The expression of miR-143 was significantly decreased and that of KRAS protein significantly increased in HL-60 cells when compared with those in peripheral blood mononuclear cells (PBMNCs) (P<0.01) .MiR-143 could specifically bind to 3'-untranslated region ( UTR ) of KRAS gene.The miR-143 expres-sion was significantly increased, the KRAS protein expression significantly reduced, and cell prolif-eration significantly inhibited in cells transfected with miR-143 mimic, which were significantly re-versed after transfection with miR-143 mimic+KRAS.Conclusion MiR-143 can inhibit the prolif-eration of acute leukemia cells HL-60 by target-controlling KRAS.
