摘要

Abstract

Objective To explore the correlation between the acute rejection (AR) in rat heart transplant (HT) with extracellular 45Ca2+ uptake rate and intracellular Ca2+ concentration of CD8+ CD28+ Tlymphocytes . Methods The flow cytometry was used to sort the peripheral blood CD8+CD28+ Tlymphocytes on postoperative 1 , 3 ,5 ,7 ,9 ,11 d in the rat control group (T0 ) ,sham operation group (T1 ) and experiment group (T2 ) ,the extracellular 45Ca2+ uptake rate and intracellular Ca2+ concentration were measured ,and then the correlation between extracellular 45Ca2+ uptake rate and intracellular Ca2+ concentration of CD8+ CD28+ Tlymphocytes was analyzed .Results The extracellular 45 Ca2+ uptake rate and intracellular Ca2+ concentration of CD8+ CD28+ Tlymphocytes in the T2 group were significantly increased compared with the T1 group and T0 group ,the difference was statistically significant(P<0 .05);the 45 Ca2+ uptake rate and intracellular Ca2+ concentration of CD8+ CD28+ Tlymphocytes in the T2 group were positively correlated with the degree of AR ,the Spearman correlation coefficients were 0 .494 and 0 .688 respec‐tively(P<0 .01) .Conclusion Peripheral blood extracellular 45Ca2+ uptake rate and intracellular Ca2+ concentration of CD8+CD28+ Tlymphocytes are closely correlated with the AR in rat HT,which is conducive to early monitor AR af ter H T.
