Abstract

Objective The objective of this study is to detect the relationship between the gas molecule hydrogen sul-fide (H2S) and oxidative stress in mice. Methods 24 male C3He mice as experimental animals were randomly divided into three groups:control group ( fed with basic food without fava beans components) , FB-treated group ( the FB-treated mice were intragastrically administered with flesh fava beans homogenate at a dose of 10 g/kg body weight with an interval of 4 hours) and H2 S-treated group ( intragastrically administered with flesh fava beans homogenate at a dose of 10 g/kg body weight, every 4 h and injected intraperitoneally with 25μM/kg hydrosulfide sodium NaHS at an interval of 4 hours) . After 24 hours of treat-ment, the content of hydrogen peroxide ( H2 O2 ) , H2 S, malondialdehyde ( MDA ) and glutathione ( GSH ) and catalase ( CAT) activity were measured in blood of all groups of mice. The mice were killed by cervical dislocation, and antioxidant gene expression level in liver, such as superoxide dismutase1 (SOD1), SOD2, CAT and glutathione reductase (GSR) were investigated. Results After 24 hours of treatment, compared with the control group, the levels of H2 O2 , MDA, and H2 S were significantly elevated (P<0. 05) in the FB-treated mice, while the activity of CAT decreased (P<0. 05). Compared with the FB-treated group, the levels of H2O2, and MDA were significantly decreased (P<0. 05), and H2S, and GSH were significantly elevated (P<0. 05), while the activity of CAT was decreased in H2S group. The QRT-PCR was performed to test oxidative stress related genes expression. After 24 hours, the expressions of glutathione peroxidase3 ( GPX3 ) , thioredoxin Peroxidase, Cu -superoxide dismutase ( CuSOD) , tumor necrosis factor-α were significantly increased in the FB-treated mice compared with that of control group (P<0. 05). The expressions of SOD1, SOD2, CAT, glutathione peroxidase1, GPX3, GSR, CuSOD, TGF-β1 were significantly increased in the H2S-treated mice compared to FB-treated mice (P<0. 05). Con-clusion The results suggest that H2 S can inhibit oxidative stress induced by fava bean ingestion by regulating the expression of oxidative stress related genes in mice.
