Effects of Pim-1 Kinase on the Differentiation of Naïve CD4+T Cells

Abstract
Objective The aim of this study was to understand more about the effects of Pim-1 kinase on the differentiation of naïve CD4+T. Methods After PIM-Inh block, the naïve CD4+T cells were induced by different cytokines. The expression of T cell master transcription factors (T-bet, ROR-γt, GATA-3, Foxp3) were measured by Western Blot. And the production of lymphocytes cytokines (IFN-γ, IL-4, TGF-β, IL-17) was determined by ELSIA. Results The expressions of T-bet, GATA-3, Foxp3 and ROR-γt pro-teins in differentiation groups of naïve CD4+T cells were significantly higher than in the controls, and so were the expressions of IL-4, TGF-β, IFN-γand IL-17 cytokines. Administration of PIM-Inh resulted in inhibition of T-bet and ROR-γt expression in a dose-dependent manner. PIM-Inh also inhibited the production of IFN-γand IL-17 in a dose-dependent manner. Conclusion These data suggested that Pim-1 may play an important role in the differentiation of naïve CD4+T cells into Th1 and Th17 cells.关键词
