Abstract
Objective:To study the effect of different fractions extracted from Radix Ophiopogonis on inflammato-ry factors in LPS-induced RAW264 .7 cells and to find its anti -inflammatory parts .Methods:The methods of reflux extraction and organic solvent extraction were performed to obtain the fractions from the extract of Ra -dix Ophiopogonis .After treating cells with different extracting fractions from Radix Ophiopogonis , the MTS as-say was used to detect cell viability .The model of RAW264 .7 cell inflammatory reaction induced by Lipopo-lysaccharide was pretreated by different fractions extracted from Radix Ophiopogonis and LPS for 24 h.At last, the concentration of NO in supernatant was analyzed by Griess reaction , and the concentration of TNF-αin supernatant was analyzed by ELISA .Results:The concentration of NO and TNF -αwas obviously inhib-ited by different fractions extracted from Radix Ophiopogonis and inflammatory factors in dose dependent man-ners .Particularly , the water extractive fraction had a greater inhibitory effect on NO secretion , and the n-bu-tyl alcohol extractive fraction had a greater inhibitory effect on the expression of TNF -α.Conclusion: Differ-ent fractions of Radix Ophiopogonis have anti -inflammatory activities .The mechanism of the anti -inflamma-tion of different fractions from Radix Ophiopogonis may be related to the inhibition of the release of NO and TNF-αfrom macrophages .The results indicate that the water and n -butyl alcohol extractive fractions have the best activities .关键词
