Abstract
Objective To study the effects of high fat and calories diet on pharmacokinetics of melatonin prolonged -release tablet in healthy Chinese subjects.Methods Ten healthy subjects were randomly as-signed to receive single oral dose of melatonin prolonged-release tablet 2 mg after overnight fasting or breakfast by a randomized crossover design. Plasma concentrations of melatonin were measured by HPLC -MS/MS. The pharmacokinetic parameters were calculated, and the effect of the high fat and calories diet on the pharmacokinetics of melatonin was evalu-ated.Results Under fasted and fed conditions, the main pharmacoki-netic parameters of melatonin were as follows:Cmaxwere (1.40 ±0.68), (1.13 ±0.38)ng・ mL-1, t1/2 were (2.53 ±1.10), (2.64 ±1.85)h, tmax were (1.28 ±0.85), (2.28 ±1.52)h, AUC0-10 h were (4.69 ±2.21), ( 4.09 ±1.58 ) ng・ mL-1 ・ h, AUC0-∞ were ( 4.98 ±2.45 ) , (4.32 ±1.64) ng・ mL-1・ h.Conclusion The absorption of melato-nin was reduced by food intake.关键词
