miRNA-16对肺腺癌A549细胞增殖和VEGF-A表达水平的影响
Effect of miRNA-16 on proliferation and expression of VEGF-A in A549 cells
摘要
Abstract
Objective To observe the effects of miRNA-16 on the proliferation and expression of VEGF-A of A549 cells. Methods Human lung adenocarcinoma A549 cells were transfected with 50 nmol/L miRNA-16 mimics or negative control with Lipofectamine 2000. 24 hours after transfection, the expression level of miRNA-16 was meas-ured using qRT-PCR. Cell proliferation was measured by methylthiazol tetrazolium( MTT) assays. The expression level of VEGF-A was tested with enzyme-linked immunosorbent assay ( ELISA ) . Results The expression level of miRNA-16 in A549 cells was significantly up-regulated compared with the blank control group and negative control group(P<0. 05). The proliferation of A549 cells was obviously inhibited(P<0. 05). The expression of VEGF-A in A549 cells was decreased in miRNA-16 mimics group conpared with other two groups(P<0. 05). Conclusion The results indicate that miRNA-16 inhibits the proliferation of A549 cells, which might be associated with the down-regulation of the reduced expression of VEGF-A.关键词
