摘要

Abstract

A glutamate dehydrogenase gene from A rachis hypogaea L .was obtained by using the in silico cloning technique .Its sequence was amplified using primers designed according to the cDNA sequence .The sequencing confirmed and named the gene AhGDH1 with a GenBank accession number of KT933119. Through the bioinformatic analysis ,the composition of amino acid sequence ,basic physical and chemical properties ,subcellular localization , transmembrane region , signal peptide , guide peptide , and secondary and tertiary structures of the protein were studied .The results showed that the length of cDNA of the gene was 1 713 bp ,which included a 1 236 bp open reading frame and encoded 411 amino acids residue .The encoding protein of the AhGDH1 gene consisted of two typical conserved domains .The homology comparison and phylogenetic analysis indicated that the amino acid encoded by AhGDH1 gene in A .hypogaeawas highly homologous with those encoded by GDH1 gene in Cicer arietinum and others . Thus , it was determined that the protein was a glutamate dehydrogenase . The information provided for the expression analysis and function identification of GDH1 gene in A .hypogaea in the future .
