一种海洋黑曲霉耐盐内切纤维素酶的分离纯化和酶学性质研究
Purification and Characterization of a Halotolerant Endoglucanase from MarineAspergilla Niger

Abstract
A marine-derivedAspergillaniger strain was used to produce cellulase by solid state fermentation. The endoglucanase was purified by ammonium sulfate precipitation, ultra-filtration, anion exchange chromatograph and hydrophobic interaction chromatography, and the molecular weight of the endoglucanase was~34 kDa. The specific activity of the purified endoglucanase increased to 557.27 U⋅mg−1 from 37.72 U⋅mg−1 of the crude endoglucanase. The results show that the optimal temperature and pH for better endoglucanase activity are 60℃ and 4.5, respectively. The endoglucanase activity retains up to 87% at 50℃ and 57% at 60℃ after 9 h, and it is stable over a broad pH range. Cu2+ and Fe2+ ions are found as inducers while Mn2+, Ni2+ and Zn2+ significantly inhibit enzyme activity. The endoglucanase activity in 20~60 g⋅L−1NaCl solutions is higher than that in NaCl-free solutions, and the residual activity of about 89% at 20% salt concentration indicates its halotolerance nature. The kinetic constantsKm andVmax determined for endoglucanase with carboxylmethyl cellulose as a substrate are 11.7 mg⋅mL−1 and 943.4 U⋅mg−1, respectively.关键词
