摘要

Abstract

The objective of the present study was to investigate into the functions that Schistosoma japonicum calreticulin protein (SjCRT) activated mouse macrophagocytes. Insect cells Sf9 were used to express SjCRT. Macrophagocytes were stimulated with SjCRT for 72 h. The expression levels of macrophagocyte surface chemokine receptor CCR7 were assayed with flow cytometry. The expression levels of COX-2, cPLA2, TNF-α and IL-1β mRNAs were assayed in RT-PCR. The ELISA was used to assay the concentration of TNF-α and Griess to assay the concentration of NO in the surpernatant samples. The flow cytometry results show that SjCRT significantly promoted up-regulation of the CCR7 expression (P<0.05) in macrophagocytes. RT-PCR results indicated that SjCRT upregulated mRNA levels of COX-2, cPLA2, TNF-α and IL-1β. According to the results of ELISA and Griess,SjCRT significantly stimulated macrophagocytes to secret NO. In brief,SjCRT activated mouse macrophagocytes that might be involved in the response to schistosomiasis vaccination.
