Effect of Surfactant Protein D on Genetic Predisposition of Patients with Allergic Rhinitis in Central China Area

Abstract

Objective To investigate the genetic etiology and characteristics of patients with allergic rhinitis in China.Methods Peripheral blood samples were collected from 216 Han adult patients with allergic rhinitis (female: 113 cases, male: 103 cases, average age:34.3 ±14.38 year-old ). Single nucleotide polymorphisms of surfactant protein D (SP-D)gene was detected by using pyrosequencing technology and mutant allele frequencies was fuetheranalysed.Files of history data in detailwere established,basing on the illnesshistory-takingand questionnaire which included family history,maternal history of sensitization and medicationduring pregnancy, history of premature delivery, living environment and petsgrooming ect.Results The frequencies of CC genotype and allele C of rs721917 site were significantly higherin allergic rhinitis group,compared to control group.The risk of genotype CC was 2.847-fold (OR =2.847,95% CI:1.313-6.176)times higherthan TT genotype,where as the risk of C allele resulted in 1.633-fold (OR =1.633,95% CI:1.153-2.397)increasing for suffering from allergic rhinitis.It suggested that rs721917 C allele could be the candidate of susceptibility geneamong Chinese Han allergic rhinitis population,based on the results of relative risk anlysis on genotype frequencies.The difference of allele frequencies of rs2243639 and rs3088308 was not statistically significant between allergic rhinitis group and control group,indicating that these two sites did not play key roles in the pathogenesis of allergic rhinitis.Conclusions Single nucleotide polymorphism of SP-D gene is associated with genetic susceptibility of allergic rhinitis in Central China area.It may be an important candidate of susceptibility gene for allergic rhinitis.The SP-D gene polymorphism may play an key role in the pathogenesis of allergic rhinitis.Met11 Thr (rs721917)is the most common polymorphism site.SP-D gene may be an important candidate of susceptibility gene for allergic rhinitis.
