量子点荧光成像技术在人舌鳞癌细胞缺氧环境下的应用研究

Abstract
Objective:To develop a QDs-based approach for HSP70 expression kinetics on human tongue cancer Tca8113 cells under hypoxia environment induced by CoCl2 in different time points. Method:The human tongue cancer Tca8113 cells were cultured in normoxic and hypoxia environment induced by CoCl2 respectively. HSP70 protein was labeled by specific fluorescent tags. And the expression of it was observed using confocal laser microscopy. The change of HSP70 fluorescence signal intensity was detected under hypoxia environment 0 h, 6 h, 12 h, 24 h, 36 h, 48 h by Image-Pro Plus software. Result: At 6 h hypoxia exposure, the synthesis of HSP70 was increased, and had a tendency to gather in the nucleus and reached the maximum value at 24 h. Conclusion:CoCl2 can lead to upregulated expression of HSP70, stimulate the proliferation of Tca8113 cells under induced hypoxia at different time points which suggesting that HSP70 maybe an important role in the development of human tongue cancers.关键词
