番茄APX基因的分离及表达分析
Isolation and expression analysis of LeAPX gene in tomato

Abstract
Imbalance of metabolism can cause increased accumulation of ROS.If it could not be scavenge timely,it will lead to damage to cellular components.In antioxidative system,Ascorbate peroxidase (APX)has been considered as pivotal enzyme for scanvenging ROS in chloroplasts and protecting cellular components.In the present study,we isola-ted an APX gene,designated LeAP X ,from tomato.The 1 412 bp cDNA contained an open reading frame (ORF)com-prising of 1 266 nucleotides (nt)which encodes a polypepetide of 421 amino acid residues.The deduced amino acid se-quence showed high identity with APXs from Solanum tuberosum ,Sesamum indicum and Nicotiana tabacum .Amino acid sequence alignment indicated that the APX contained two domains.Sequence analysis of the promoter indicates that it is rich in acting elements such as HSE、LTR、GT1. qPCR indicated that the gene showed the highest expression level in leaves comparing with other organs.关键词
