薏苡仁不同组分对脾虚水湿不化大鼠空肠基底膜相关基因的影响
Effect of different components of Coix Seed on related genes in jejunal basolateral membrane of rats with syndrome of spleen deficiency and wa-ter dampness
摘要
Abstract
Objective To investigate the effects of different components of Coix Seed(Semen Coicis, Yiyiren)on certain genes of transporters and factors in jejuna basolateral membrane of rat with syndrome of spleendeficiency and wate rdampness.Methods Aftermodeling,theratswithsyndromeofspleen deficiency and water dampness were treated by different components extracted from Coix Seed.Then jejunum was tested by gene chip;functional-annotation-gene to basolateral plasma membrane was analyzed by Quick-GO,and the most representative gene was found out by principal component (PC )analysis.Results Therewere 20 differential genes in sevengroups.After dimension reduction by principal component analysis (PCA ),89.7% of variance could be explained when the number of principal component was three.By correlation analysis between loading and score in PC1 and PC2,the expression of genes on basolateral plasma membrane of PC1 was very close to those of PC2,showing the similar function of different components of Coix Seed on jejuna basolateral membrane.However,there was a different situation in PC2 and PC3.Normalization of the weight of PC1,PC2 and PC3 showed that the leading weight genes were Slc22a7,Car9,Agtr1b,S100g,Aqp9,Msn,Aqp3,Slc4a1,and Slc4a10.Conclusion Coix Seed can repair the injury of small intestine due to modeling stimulation, and upregulate SLC expression leading to regain the ability of raising white blood cell,materializing the TCM abstract term “invigorating spleen”.Coix Seed can downregulate AQP3 expression of the model, and reduce liquid absorption from intestinal lumen,materializing the term of“eliminating dampness”.关键词
