响应曲面法优化野蔷薇根多糖的提取工艺及抑菌作用研究
Response Surface Methodology for Optimization of Extraction Conditions of Polysaccharide from Rosa Multiflora Thunb and Antibacterial Activity
摘要
Abstract
To optimize the extraction technology of Rosa Multiflora Thunb polysaccharides by response surface method(RSM)and test its antibacterial activity .Extraction temperature,microwave power,extraction time and ratio of material to liquid were investigated by single factor test and RSM using the yield of Rosa Multiflora Thunb poly-saccharides as index to optimize the extraction technology. The antibacterial activity was evaluated by the methods of disk diffusion and standard tube dilution. The optimal condition obtained from RSM was as follow:extraction tem-perature 81℃,microwave power 317W, extraction time 22 min,the ratio of liquid to material 32:1(mL/g). The yield of polysaccharides was 6.10mg/g,which was in line with theoretical value basically . The polysaccharide from the Rosa Multiflora Thunb had strong inhibiting effects on some pathogenic bacterium such as Bacillus subtilis, Escherichia coli、Staphylococcus aureus and Proteus mirabilis. The model well predict the yield of Rosa Multiflora Thunb polysaccharides,and RSM is feasible to optimize the extraction technology of RosaMultifloraThunb polysac-charides.关键词
