水貂白细胞介素－6基因的克隆与序列分析
Cloning and Sequence Analysis of Mink IL -6 Gene
摘要
Abstract
One pair of primers specific to mink Interleukin -6(IL-6) gene were designed and synthesized according to the pig IL -6 sequence in GenBank ,and were used to amplify mink IL-6 cDNA by RT -PCR from total RNA of mink peripheral blood lymphocyte induced with Phy-tohemagglutinin(PHA) ,RT-PCR product was cloned into pMD18-T vector .The nucleotide acid sequence had been determined after being i-dentified by restriction endonucleases digestion .The result showed that the cloned mink IL -6 cDNA was 633bp in length with a 633bp open reading frame and encoded a polypeptide of 210 amino acid .Compared with published domestic ferret IL -6 ,nucleotide acid and amino acid homology are 98.4% and 96.7% ,respectively .The cloning of the IL-6 gene provided a basis for further investigation of the biological func-tion of mink IL-6 .关键词
