摘要

Abstract

Objective:To discuss recombinant human interferon (IFNα-2b) on human chronic myelogenous leukemia cell acute transformation phase cell line K562 cell proliferation and focal adhesion kinase (FAK) mRNA expression.Method:The inhibitory rate of IFNα-2b on K562 cells was detected by MTT method and the effect ofα-2b on the expression level of mRNA FAK in K562 cells was detected by RT-PCR technique. Result:In addition to 48 h and 72 h,the ratio of inhibiting K562 cell proliferation increased gradually by increasing IFNα-2b concentration and prolonging reaction time.IFNα-2b in 48 h and the concentration 10 thousands U/mL was the highest to inhibit cell growth.With the increase of the IFNα-2b concentration,FAK mRNA expression was increased by K562 cell,the highest expression level was experimental FAK mRNA of 10 thousands U/mL IFNα-2b,the differences were statistically significant of each groups(P<0.05). Conclusion:The abnormal expression of FAK mRNA in leukemia K562 cells,IFNα-2b may inhibit the proliferation of K562 cells by up regulating the expression level of normal mRNA FAK.
