Abstract
OBJECTIVE:To establish the content determination method of apigenin , quercetin and luteolin in the leaves of Exochorda serratifolia S.Moore..METHODS:Acid hydrolyzation was adopted, and the acid concentration, hydrolysis temperature , hydrolysis time were investigated and contents of the flavonoid aglycones were determined by HPLC .The Promosil C18 (4.6 mm ×250 mm, 5 μm) column was adopted, the mobile phase was consisted of acetonitrile-2%glacial acetic acid( V∶V=20∶80), the column temperature was 25 at sample size of 10 μl and flow rate of 1.0 ml/min.The detection wavelength was set at 267 nm.RESULTS:The conditions of acid hydrolysis were:concentration of hydrochloric acid at 4.0 mol/L with temperature at 80℃, for 5 h.The calibration curves were linear in the range of 0.82-8.20, 0.869 6-8.696 0 μg and 0.777 6-7.776 0μg.The average recovery rates were 95.8%,97.1%and 99.6%, respectively .The precision , stability and repeatability of content determination conform to the related regulations .CONCLUSIONS:This method is accurate and reliable and can be used for the quality control of leaves of Exochorda serratifolia S.Moore..关键词
