siRNA 沉默胸腺素β4对鹿茸间充质干细胞增殖的影响

摘要
Abstract
Objective To investigate the effects of siRNA-mediated thymosin β4(Thymosin β4 ,Tβ4 )silence on proliferation in activated mesenchymal stem cell(mesenchymal stem cell,MSC)and discuss the possible mechanism. Methods MSC cells were divided into 2 groups:negative control group and siRNA-Tβ4 transfection group. The siR-NA-Tβ4 was transfected into MSC cells and cell proliferation was measured by MTT,Real-time PCR detection ILK, AKT and CyclinD1mRNA expression quantity change,Western blot detection AKT and P-AKT protein content chan-ges. Results MSC proliferation was significantly inhibited in siRNA-Tβ4 transfection group(P < 0. 01);Expression of the ILK,CyclinD1 mRNA of MSC transfected with siRNA-Tβ4 is reduced,AKT quantity remains the same( P <0. 01);The siRNA-Tβ4 transfection group no change in total AKT protein,P-AKT protein content decreased. Conclu-sion The siRNA-mediated Tβ4 silence significantly inhibits MSC proliferation by up regulating the ILK,p-AKT and CyclinD1expression in MSC.关键词
