摘要
Abstract
Objective To establish an HPLC method to determine the concentration of theophylline in human plasma. Methods The serum sample was performed the precipitation treatment by 10% trifluoroacetic acid , the chromatographic column was the Inertsil ODS-SP column ( 250 mm × 4. 6 mm, 5 μm ) , the mobile phase was consisted of methanol-water ( 25 :75 ) . The UV detection wavelength was at 274 nm with a flow rate of 1. 0 mL/min and the column temperature of 25 ℃. Results The concentration of theophylline in the range of 2. 55-40. 72 μg/mL showed good linearity. The regression equation of plasma theophylline concentration and peak area was Y=( 45 417. 2 ) X-4 580. 0, r=0. 999 8 ( n=6 ) . The intra-day and inter-day RSD were less than 4. 88% and 6. 06%, and the limit quantitation was 0. 2 μg/mL. Conclusion The established method is fast, accurate and suitable for therapeutic dug monitoring ( TDM ) of theophylline in clinical.关键词
