PA-MSHA对脓毒症/严重脓毒症大鼠肠道ICAM-1、VCAM-1表达的影响
Effect of PA-MSHA on expression of ICAM-1 and VCAM-1 of intestinal tract in septic/severe septic rats

Abstract
Objective To study the effect of pre-administration of pseudomonas aeruginosa-mannose sensitive hemagglutinin (PA-MSHA) on the expression of intestinal intercellular cell adhesionmolecule-1 (ICAM-1) and vascular cell adhesion molecule 1 (VCAM-1)in septic/severe septic rats.Methods 40 healthy SPF rats were randomly divided into five groups: blank group, the septic group and the severe septic group were given normal saline subcutaneous injection of 0.3 ml/day and PA-septic group and the PA-severe septic group were given PA-MSHA subcutaneous injection of 0.3 ml/d for 7 consecutive days. At the 8th day, five groups underwent intraperitoneal injection of the modified Gonnert human stool suspension to make septic/severe septic rat models. After monitoring vital signs, intestinal tissue from five groups was harvested and analyzed with HE staining and enzyme-linked immunosorbent assay (ELISA) to detect the expression of ICAM-1 and VCAM-1 in the intestinal tissue of rats.Results (1) HE staining showed that the morphological changes compared with blank group; (2) The average expression of ICAM-1 (F=33.337,P<0.001) and VCAM-1 (F=12.714,P<0.001) in intestinal tract shown statistically significant differences of five groups; (3) The expression of ICAM-1 was increased in septic group compared with PA-sepsis group (P<0.05), and severe septic group was increased PA-MSHA group (P<0.05). (3) The expression of VCAM-1 was increased in sepsis group compared with PA-sepsis group (P<0.05), and there was no significant difference in the expression of VCAM-1 between the severe septic group and PA-severe sepsis group (P=0.262).Conclusions (1) The expression of ICAM-1 and VCAM-1 in intestinal tract in septic/severe septic rats was increased. (2) PA-MSHA pretreatment can inhibit the expression of ICAM-1 and VCAM-1 in intestinal tract in septic group. (3) In severe septic rats, the level of ICAM-1 and VCAM-1 can not accurately reflect the effect of PA-MSHA. The reason may be related to necrosis of intestinal epithelial cells and inflammatory cells.关键词
