双氢杨梅树皮素对人视网膜母细胞瘤HXO-RB44细胞色素C氧化酶表达的影响
Influence of ampelopsin on expression of cytochrome C oxidase of human retinoblastoma HXO-RB44 cells

Abstract
Objective To explore the influence of ampelopsin( APS) on the expression of cytochrome C oxidase IV( COX-IV) of human retinoblastoma( RB) cell cultured in vitro.Methods Human retinoblastoma RB cell lines( HXO-RB44) cultured in vitro were divided into three groups according the APS concentration,group A (4.40μg/ml),group B(14.84μg/ml) and group C(33.33μg/ml). The blank control group and positive control group(cisplatin,2.00 μg/ml) were enrolled in the study.The COX-IV expression was determined in each group by Western Blot.Results The COX-IV expression decreased significantly in each APS group after RB cells cultured in vitro had been treated for 24 hours, which showed a significant difference among each APS group, blank control group and positive control group(P<0.05).The decrease of COX-IV activity expression was the most significant in group A,and the relative grey value was (0.112 ±0.032).No group presented a characteristic of higher drug concentration and lower COX-IV activity expression.Conclusion APS can reduce the expression of COX-IV.The COX-IV expression may own the threshold,and its expression doesn′t decrease with the increasing APS concentration.关键词
