泡桐叶绿体游离及其DNA提取方法的筛选
Screening on Extraction Method of Paulownia Chloroplast DNA
摘要
Abstract
To further study the Paulowniachloroplast genome ,the Paulownia fortunei seedlings derived from tissue culture were used as test material to extract the chloroplast DNA by three methods :sucrose density gradient centrifugation ,differential centrifugation and high-salt low-pH method ,the mass con-centration of w hich w as 92 .1 ng/μL ,968 .9 ng/μL and 175 .9 ng/μL respectively .T he results of micro-scopic observation and agar gel electrophoresis indicated that the best method to obtain the pure cpDNA was to extract the gross chloroplast firstly by differential centrifugation ,then purify the gross chloroplast by sucrose density gradient centrifugation ,and finally get pure cpDNA by SDS method ,which could meet the requirements of chloroplast genome research .关键词
