Abstract
Objective:To observe the effect of reduced glutathione on oxidative stress in the rodent model of cisplatin induced acute kidney injury(AKI). Method:66 male wistar rats were randomly assigned to three groups.The rats of CP group(n=30) were injected once intraperitoneal with cisplatin at 10 mg/kg, the rats of CN group were injected intraperitoneal with same volume of normal saline vehicle,the GI group were injected intraperitoneal with reduced glutathione at 800 mg/kg before cisplatin injection.Rats were euthanized at 12,24,48,96 and 144 hours(n=6 in the CP group at each time point and n=6 in the GI group at each time point).The blood and kidney samples were analyzed at each time point.Result:A significant increase in serum creatinine was noted by 48 hours in the CP group versus CN group,peak levels of serum creatinine were seen on 144 hours(P<0.05).The levels of MDA in renal tissues were significantly higher by 48 hours in the CP group than in the CN group(P<0.05).The levels of SOD in renal tissues were significantly lower by 48 hours in the CP group than in the CN group(P<0.05).Compared with CP group,the levels of SOD in renal tissues increase significantly in GI 48 hours group,while the levels of MDA in renal tissues decreased significantly in GI 48 hours group.Conclusion:Reduced glutathione can attenuate the cisplatin induced AKI in rodent model,which may be associated with anti-oxidative stress effect.关键词
