高渗盐羟乙基淀粉溶液联合己酮可可碱对重度失血性休克大鼠再灌注后肝脏的影响
Effects of hypertonic saline/hydroxyethyl starch solution combined with pentoxifylline on the livers of rats with severe hemorrhagic shock after reperfusion

Abstract
Objective:To observe the effects of hypertonic saline/hydroxyethyl starch solution (HSH)combined with pentoxifylline (PTX) on the livers of rats with severe hemorrhagic shock after reperfusion and to reveal the underlying mechanisms.Methods:Seventy-two Sprague-Dawley rats were randomly divided into 6 groups,including control group (group C),shock group (group NR),lactated Ringer's solution-treated group (group LR),LR solution and PTX group (group LR/PTX),HSH-treated group(group HSH),HSH and PTX group (group HSH/PTX group),with 12 rats in each group.The hemorrhagic shock model was made through improved Wigger' s method.The contents of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in blood plasma,the expression of endothelin-1 (ET-1),nuclear factor-κB (NF-κB),tumor necrosis factor-α (TNF-α),MIP-2 and myeloperoxidase (MPO) activity in liver tissue were detected.Morphological changes of liver were observed under the light and electron microscopes.Results:Compared with group LR or HSH,the content of AST and ALT in blood plasma,expression of NF-κB,ET-1,TNF-α,MIP-2 and MPO activity in liver tissue were significantly decreased in group LR/PTX and HSH/PTX (P < 0.05).Liver injury was obviously less especially in group HSH/PTX.Conclusion:Liver injury can be well protected by HSH/PTX probably through reducing the production of ET-1,TNF-α and MIP-2,suppressing the activation of NF-κB,and reducing the activity of MPO in rats with severe hemorrhagic shock after perfusion.关键词
