摘要

Abstract

AIM:To investigate the effects and mechanism of anti-digoxin antiserum on the apoptosis induced by ischemia/reperfusion (I/R) in neonatal cardiomyocytes.METHODS:Primary neonatal rat cardiomyocytes were incubated in ischemia mimetic solution for 2 h,and then reincubated in normal culture medium for 24 h to establish a model of ischemia/reperfusion.The cardiomyocytes were randomly divided into:control group,I/R group,Oua + I/R group,and DigAb + I/R group.Cardiomyocyte apoptosis was detected by Annexin V-FITC/PI.The expressions of endoplasmic reticulum stress associated protein GRP78 and CHOP were analyzed by Western blot.RESULTS:Compared with the control group,apoptosis cardiomyocytes were significantly increased in I/R and Oua + I/R groups (P < 0.001).Treatment of anti-digoxin antiserum decreased I/R-induced apoptosis cardiomyocytes (P < 0.001).The expressions of GRP78 and CHOP were significantly increased in I/R and Oua + I/R groups (P < 0.01).The expressions of GRP78 and CHOP were inhibited by treatment of anti-digoxin antiserum (P < 0.05).CONCLUSION:Anti-digoxin antiserum decrease cardiomyocyte apoptosis induced by I/R,and its mechanism may be partly by inhibiting endoplasmic reticulum stress.
