摘要

Abstract

AIM:To investigate the effects of berberine on the gene mRNA expression of FGF21/SIRT1/PRDM16 signal pathway and PRDM16 signal pathway in visceral white adipose tissues (VWAT) from obesity-related type 2 diabetic Chinese hamsters and the related mechanisms.METHODS:The obese insulin-resistant hamster models were induced by high-fat diet and type 2 diabetic hamster models were induced by obese insulin-resistant hamster models injected with low-dose streptozotocin.After that,the hamsters were randomly divided into control,obese insulin-resistant,obese type 2 diabetic and berberine-treated type 2 diabetic groups.After nine-week berberine treatment,the gene mRNA expression changes of VWAT FGF21/SIRT1/PRDM16 signal pathway and PRDM16 signal pathway from different groups were measured by using real-time RT-PCR.RESULTS:The gene mRNA expression of FGF21,FGFR1,βKL,SIRT1,GLUT-1,PRDM16,CtBP-1,CtBP-2,C/EBPβ,PPARγ,PGC-1α,PGC-1β and brown adipose tissue-specific genes such as UCP-1,Cidea,Elovl3 and PPARα was decreased and that of white adipose tissue-specific genes including Resistin,MEST and Serpina3k was increased in dysfunctional VWAT from hamster models.Berberine treatment induced VWAT FGF21/SIRT1/PRDM16 signal pathway and PRDM16 signal pathway,induced the gene mRNA expression of brown adipose tissue-specific genes and inhibited that of white adipose tissue-specific genes,induced browning of white adipose tissues and improved fat-induced insulin resistance.CONCLUSION:FGF21/SIRT1/PRDM16 signal pathway and PRDM16 signal pathway involved in the inductive molecular mechanisms of berberine on browning of visceral white adipose tissues in type 2 diabetic hamsters.
