摘要

Abstract

Objective To investigate whether exenatide ( EXE ) re-duced palmitic acid induced hepatocyte apoptosis via inhibiting endoplas-mic reticulum stress.Methods Human liver L-02 cells were divided into six groups:control, model (treated with palmitic acid 500μmol· L-1 for 24 h), test-L (treated with palmitic acid 500μmol· L-1 and EXE 25 nmol· L-1 for 24 h), test-M (treated with palmitic acid 500μmol· L-1 and EXE 50 nmol· L-1 for 24 h) , test-H ( treated with palmitic acid 500 μmol · L-1 and EXE 100 nmol · L-1 for 24 h ) and combination ( treated with palmitic acid 500 μmol · L-1 , EXE 100 nmol · L-1 and tunicamycin 2 μg· L-1 for 24 h).Cell vitality was determined by 3-(4,5-dimethyl -2-thiazolyl) -2,5-diphenyl -2-H-tetrazolium bromide (MTT).TdT-mediated dUTP nick -end labeling ( TUNEL) staining and Caspase -3 activity assay was used to detect cell apoptosis.For endoplasmic reticulum stress detecting , the expression of C/EBP ho-mologous protein ( CHOP) was analyzed by real -time polymerase chain reaction ( RT -PCR ) . The Caspase -12 activity was detected&nbsp;by Caspase-12 activity assay kit.Results EXE dose -dependently increased cell vitality , decreased apoptotic in-dex, Caspase-3 and 12 activity, as well as down -regulated CHOP expression in palmitic acid treated L -02 cells ( all P<0.05 ).As compared to test -H group, CHOP expression , apoptotic index and Caspase -12 activity were increased , while cell vitality were decreased in combination group [ ( 78.58 ±1.78 )% vs ( 87.78 ±3.66 )%] ( all P<0.05 ).Conclusion EXE may reduce fatty acid induced hepatocyte apoptosis through inhibiting endoplasmic reticulum stress.
