Abstract

AIM To develop a method for the determination of mitoxantrone in rat plasma by HPLC.METHODS The plasma sample preparation was precipitated with 10% trichloroacetic acid-methanol solution.Chromatographic separation was carried out on a Xbridge-C18 column (150 mm× 2.1 mam,3.5 μm) with mobile phase consisted of 0.3 %sodium heptanesulfonate solution (pH was adjusted to 2.0 with phosphoric acid) and acetonitrile (72:28).The flow rate was 1.0 mL· main-1,and the detection wavelength was 240 nm.Bupropion was used as internal standard.The established method was applied to pharmacokinetic study of mitoxantrone in rats.RESULTS Excellent linear calibration curve of mitoxantrone was obtained in the plasma concentration range of 100-10 000 μg· L-1 (r =0.999 6).The relative standard deviations of inter-and intra-day precisions were all less than 6%.The average recovery was above 80%.The relative standard deviations of short-term room temperature preservation and long-term cryopreservation stabilities were all less than 8%.Dosage of 5 mg· kg-1 mitoxantrone was given to rats by intravenous injection,its AUC(0→t) was (1 178.44±70.07) μg·h·L-1,t1/2z was (0.39±0.12) h,CL z/F was (3.91 ±0.26) L-1·h·kg-1,Vz was (2.18 ± 0.63) L· kg-1,tmax was (0.22 ± 0.04) h and ρmax was (2 380.91 ± 360.12) μg· L-1 respectively.CONCLUSION The method is simple,specific and reliable for the pharmacokinetic research of mitoxantrone in rats.
