Analysis of the specific effects of long chain non encoding RNAMEG3 in the proliferation of gastric cancer cells

Abstract
Objective To analyze the long non-coding RNA MEG3 expression levels in gastric cancer and explore MEG3 specific impact on gastric cancer cell proliferation by over-expression.MethodsThe normal gastric tissues and cells as the control group, the expression levels of gastric carcinoma cells and MEG3 as the observation group, using qRT-PCR(quantitative reverse transcription PCR) technique MEG3 expression levels in the observation group were detected using transfection pcDNA-MEG3 be raised, to detect the transfection efficiency. Then after impact MEG3 raise the level of proliferation of BGC-823 and MGC-803 cells using MTT assay testing, and by Western blot detection of two cells p53 protein levels.Results (1)measuring 36 cases of gastric cancer tissues and the corresponding normal tissues adjacent to cancer tissue groups in the presence of water on average relative expression MEG3 significant difference was statistically significant(P<0.05) with fluorescence quantitative PCR. (2)MGC-803 and BGC-823 cells were transfected with pcDNA-MEG3, compared with a significant increase in the level of the control group MEG3 310 and 251 times the difference between the groups was significantly(P<0.05). (3) By MTT assay, after rising MEG3 level, proliferation MGC-803 and BGC-823 cells was significantly decreased(P<0.05). (4)And the control group, MGC-803 and BGC-823 transfected pcDNA-MEG3, which significantly increased the expression of p53(P<0.05). Conclusion The gastric cancer tissues and cells MEG3 decline can inhibit the activation of p53 protein, leading to gastric cancer cell proliferation and the development process of gastric cancer.关键词
