Abstract

Objective] To research the multiplication subculture technology of three clones of Eucalyptus urophylla × E. grandis, and to pro-vide theoretical foundation for the rapid propagation of E. urophylla × E. grandis seedlings. [ Method] With three clones (DH32-26, DH32-29, DH32-28) as the test materials, the optimal combination of regulators was screened, as well as the coagulator, dark culture days and culture flask. Thus, multiplication coefficient was relatively high. [Result] The optimal combination of plant growth regulators for subculture of three clones were DH32-26(6-BA 0. 5 mg/L + NAA 0. 2 mg/L), DH32-28(6-BA 0. 4 mg/L + NAA 0. 2 mg/L) and DH32-29(6-BA 0. 3 mg/L +NAA 0. 2 mg/L). DH32-26 multiplication coefficient showed no significant differences in culture medium with carrageenan and agar as the coag-ulators. Subculture multiplication coefficients of DH32-28 and DH32-29 in culture medium with carrageenan as the coagulator was higher than those with agar as the coagulator. During the process of rooting, culture medium with carrageenan as the coagulator had far better rooting than that with agar as the coagulator. In the initial stage of DH32-26 and DH32-28 subculture, black cloth was used for shading for 5-10 d; while DH32-29 did not need all dark culture. DH32-29 subculture in wild-mouth bottle reduced the production cost of seedlings; but DH32-26 and DH32-28 were not suitable for the subculture in wild-mouth bottle. [Conclusion] To enhance the multiplication coefficient of clones, we should select the optimal regulators, coagulator, dark culture days and culture flask.
