Detection and Comparison of DNA Content in Fruit Flies in Different Periods
摘要
Abstract
Objective] The aim was to determine DNA content in 8 fruit flies at different ages.[Method] Total DNA was extracted from Ⅰ,Ⅱ, Ⅲ instar larva, pupa, imago of 7 mutant lines of Drosophila virilis and Drosophila melanogaster.DNA content was determined by ultravio-let visible spectrophotometer, PCR detection was conducted on DNA.[Result] The contents of DNA in eight fruit flies I, II and III instar lar-va were increased, and the contents of DNA in most of the pupa and imago were decreased;the efficiency of PCR of fruit flies in five periods were all good, imago was the best, larva was better.[ Conclusion] The DNA quality of larva, pupa and imago all can ensure the experiments carry out such as the PCR amplification, molecular cloning, species identification and so on.关键词
