Abstract

Objective To investigate the expression profiles of multi-drug resistance protein 1(MDR1) in the human cornea , C57BL/6 mouse cornea ,STZ induced diabetic C57BL/6 mouse cornea .Methods In the presented studies ,PCR ,western Blot-ting ,and immunohistochemistry/immunofluorescence were used to obtain the relative mRNA and protein expression profiles of MDR1 in the human cornea ,human corneal endothelial cell line(HCEC) ,C57BL/6 mouse cornea ,and C57BL/6 mouse cornea with STZ induced type Ⅰ diabetes mellitus .Results There was just(9.11 ± 2.33)Qty · ng -1 RNA of MDR1 expression pro-files in mRNA levels in human cornea ,and no expression of MDR1 in HCEC in mRNA levels ,but MDR1 had relatively high level of MDR1gene expression profiles in C57BL/6 mouse .The MDR1 expression was lowered in C57BL/6 mouse cornea with STZ induced type Ⅰ diabetes mellitus when it compared that of the normal mouse at 4 ,12 and 24 weeks disease model .West-ern blotting results showed that ,only faint MDR1 expression was observed in the human cornea ,but it was obviously observed in the HCEC ,the expression changing profiles were similar to the mRNA levels to the normal and diabetic mice .To the expres-sion sites ,MDR1 was detected in the whole human corneal epithelium ,and it was in the whole cytoplasm in HCEC ,the results of immunofluorescence also showed the the similarily expression changing profiles to the mRNA levels to the normal and diabet-ic mice .Conclusion Evidence of cross specice MDR1 expression differences noted in this study supports the conclusion that MDR1 expression variability should be taken into consideration to better understand ocular pharmacokinetic and pharmacodyna-mic data .This work also shed light on the potential limitations of animals in predicting the corneal permeability of ophthalmic drugs and transporter substrates .
