Abstract

The entire mitochondrial genome of Odontobutis was obtained by PCR based on conservative primers. It was showed that the mitochondrial genome sequence was 16, 930 bp long, including 13 protein genes, 22 tRNA genes, 2 rRNA genes and a noncoding region. The base composition of the genome was A 30.0%, C 28.9%, G 15.6% and T 25.5%. Protein-coding genes in Odontobutis mitochondrial genome used ATG as the ini⁃tiation codons with the exception of CO1 which used GTG and ND6 used TAA. It was found that stop codons in Odontobutis included TAA, uncomplement codons TA and T. Most tRNA genes could form typical secondary structures except tRNASer(AGN) and tRNACys. In addition, the D-loop region of Odontobutis mtDNA was 844 bp in length. Compared with the mitochondrial DNA control region from other species, the extended terminal associat⁃ed sequences, central conserved sequence blocks, conserved sequence blocks and putative promoters of the mito⁃chondrial DNA control region in Odontobutis were observed. Sequence alignment of Cyt b genes of Odontobutis and other 10 species from GenBank showed that Odontobutis obscara and Odontobutis had the closest genetic re⁃lationship in the 11 species. A phylogenetic tree constructed using the Neighbor-Joining method indicated that the relationship among Sleeper Branch was close, the next were Sculpin Branch and Sciaenidae, and Cyprinid was distantly related.
