Abstract
Objective: To observe the expressions of connexin40 during the acute lung injury model of C57BL/6 mice and explore the functional correlation between connexin and lung vascular permeability . Methods: C57BL/6 mice were intraperitoneally treated with lipopolysaccharide (LPS,0.01 mg/g) for 24 hours, and then the expressions of connexin40 mRNA and protein levels in lung were measured by real -time PCR and Western-blotting or Fluorescence immunohistology , respectively.To explore correlation between connexin40 and lung vascular permeability,weight gains in isolated perfused lungs at baseline and after bo -lus infusion of platelet-activating factor (PAF) in the absence (PAF only) or presence (both PAF and gap27 40 )of gap27 40 (0.02 mg/mL) were measured by PLUGSYS system, respectively.Results: Real-time PCR and Western-blotting analysis showed that , the expressions of connexin40 mRNA and protein decreased significantly in LPS group compared with control group , which was confirmed by fluorescence imaging .Per-fusion of mouse lungs with PAF caused severe edema , but application of gap27 40 (connexin40-specific in-hibitory mimetic peptide) to isolated perfused mouse lungs reduced the extent of PAF -induced edema.Con-clusion: The increased lung vascular permeability induced by inflammatory conditions may be amplified by connexin40 despite of its decreased expression .关键词
