PDX-1在人脂肪间充质干细胞分化为胰岛分泌细胞中的作用
Effect of PDX gene 1 in human adipose-derived mesenchymaI stem ceIIs differentiating into insuIin-secreting ceIIs

Abstract
Objective To invesitigate the effect of PDX gene 1 (PDX-1 )in human adipose-derived mesenchymal stem cells (ADSC)differentiating into insulin-secreting cells.Methods ADSC after cryopreservation and anabiosis were selected and then were divided into the experimental group and control group.In the experimental group,transfering PDX-1 gene into human ADSC by using the skill of adenoviral transfection method,and the control group without the adenovirus infection.The gene transfection results were detected by Western blotting,the amount of insulin secretion in the transfected cells was detected by ELISA,the expression of Ngn3 and Islet-1 was detected by RT-PCR in the two groups.Results Fif-teen days after transfection,the insulin secretion under hypoglycemia of the experimental group and the control group was (6.98 ±0.74)and (6.98 ±0.74)ng/mL,respectively (P >0.05 );the insulin secretion under hyperglycemia was (13.38 ±0.62)and (6.50 ±0.66)ng/mL,respectively;the expression of Islet-1 in the experimental group and the con-trol group was 5.150 ±0.177 and 5.150 ±0.177,respectively;and the expression of Ngn3 in the experimental group and the control group was 5.819 ±1.629 and 1.000 ±0.120,respectively.The insulin secretion under hyperglycemia,and ex-pression of Islet-1 and Ngn3 was higher in the experimental group as compared with that of the control group,and the differ-ence was significant (all P <0.05).Conclusion PDX-1 gene plays a positive role in promoting the differentiation of hu-man ADSC into islet-like cells.关键词
