Effects of 5-HT2C receptors on the firing activity of the glutamate neurons in the lateral habenular nucleus of the hemiparkinsonian rats

摘要

Abstract

To study the changes in firing activity of the glutamate neurons and the responses of the neurons to 5-hydroxytryptamine 2C (5-HT2C ) receptors stimulation in the lateral habenualr nucleus (LHb)in rats with unilateral 6-hydroxydopamine (6-OHDA)lesions of the substantia nigra pars compacta (SNc ), and clarify the role of the LHb and 5-HT2C receptors in the mechanism of PD,the firing activities of glutamate neurons in the LHb of sham-operated and the 6-OHDA-lesioned rats were assessed by electrophysiological study.The results showed that unilateral lesions of the SNc in rats led to a more bursting-firing activity of LHb glutamate neurons (P<0.05).Intra-LHb injection of Ro60-0175 as agonist of 5-HT2C receptors resulted in increase in the firing rate of LHb glutamate neurons (P<0.05)and the percentage of burst-firing neurons (P<0.05)in sham-operated and the 6-OHDA-lesioned rats.Compared to sham-operated rats,the duration of the excitatory effect produced by Ro60-0175 was markedly longer in the&nbsp;lesioned rats.These findings indicate that the changes in firing activity of glutamate neurons may be involved in the parkinsonian-related depression.And 5-HT2C receptors are involved in the regulation of the firing activity of glutamate neurons in the LHb.
