Fbxw7 modulates 5-fluorouracil resistance of colorectal cancer

Abstract
Objectives To identify the effects of Fbxw7 expression on 5-fluorouracil (FU) resistance of colorectal cancer. Methods Immunohistochemical staining was used to detect the expression of Fbxw7 in cancer tissues from 53 patients with colorectal cancer. The association of tumor recurrence with Fbxw7 expression was analysed in the postoperative patients after 5-FU chemotherapy. shRNA was transfected into colon cancer cells HCT116 to downregulate Fbxw7 and detect the growth of cells after 5-FU treatment by CCK-8 assay and plate cloning assay. Results The expression of Fbxw7 was positive in 20.0%in the cases with tumor recurrence and 53.6%without recurrence. It was found that Fbxw7 expression correlated with postoperative tumor recurrence. 5-FU treatment reduced the mRNA and protein expression of Fbxw7 of colon cancer cells. Moreover, compared to control group, downregulation of Fbxw7 increased relative cell viability and proliferation of the cells after 5-FU treatment. Conclusions Downregulation of Fbxw7 might boost the 5-FU resistance of colon cancer cells.关键词
