Abstract
Objective:To investigate the relationship between miRNA-17 ~92 cluster and the paclitaxel chemoresistance of ovarian cancer,provided a theoretical basis for the che-moresistance of ovarian cancer. Methods:Using liposome method transfected pEGFP-N1-miR-NA-17~92 plasmid into ovarian cancer SKOV3 cells. The expressions of miR-17 ~92,BIM, PTEN were detected using Real time-polymerase chain reaction( Real time-PCR) and Western blot. Results:Compared with the SKOV3 cells, empty plasmid pEGFP-SKOV3-pEGFP-N1 transfection cell miR-17~92 content was 0. 970,there was no statistically significant difference compared with control group ( P>0 . 05 ) . The miR-17~92 levels of SKOV3-pEGFP-N1-miR-17~92 cells was 2. 082,and compared with the control group difference was statistically signifi-cant (P<0. 05). In SKOV3-pEGFP-N1-miRNA-17~92 cells and SKOV3-pEGFP-N1 cells,the differences of miR-17~92 mRNA levels were statistically significant(P<0. 05). Overexpression miRNA-17 ~92 decreased BIM protein expression, the expression of PTEN protein of the effect was not obvious. Conclusions:miRNA-17~92 gene clusters involved in paclitaxel resist-ance of ovarian cancer SKOV3 cells. The mechanism of miRNA-17~92 gene cluster affecting o-varian cancer drug resistance is through cuting BIM protein instead of PTEN protein.关键词
