摘要

Abstract

Objective To investigate the effect of acid sensing ion channels (ASICs) blocker amiloride precondition on acute gastric mucosal lesion(AGML) rat and to study the relation between AGML pathogenesis with ASICs. Methods Thirty Wistar rats were selected and randomly divided to 3 groups(10 cases per group),including the normal blank control group(NC),stress model group(WS) and blocker group(amiloride precondition group,AM). The AGML model was induced by water immersion re-straint stress(WIRS). The gastric tissues in general,gastric mucosal lesion degree,histological change under microscope and ulcer index were observed,the levels of serum NO and MPO in gastric mucosal tissue were observed at 6 h after stress. Results The gastric mucosa in the WS group showed dark red with a large of punctate hemorrhage,linear hemorrhage or flake erosion,the sub-mucosal blood vessel and dermatomal blood vessel were congested ,the epithelial cell mucous layer was thin and deleted ,the intra-cellular space was increased with different degrees of necrosis;but the gastric mucosal hemorrhage or erosion degree after 6 h of WIRS in the AM group was obviously improved compared with that in the WS group;a large number of blood capillary congestion and expansion were seen under microscope ,the epithelial cell mucosal layer had a little defects ,but was basically intact. Com-pared with the group NC,the gastric mucosal UI after 6 h of WIRS in the WS group was significantly increased,serum NO en-hanced and gastric tissue MPO level was significantly elevated,the differences were statistically significant(P<0.01);compared with the WS group,gastric mucosal UI,serum NO level and gastric tissue MPO level were significantly decreased,the differences were statistically significant(P<0.01). Conclusion Amiloride can effectively alleviate the WIRS rat acute gastric mucosal lesion mediated by inflammation,which prompting that ASICs may participate in the occurrence of AGML in WIRS rat.
