Abstract

Objective To investigate the mechanism of tetramethylpyrazine (TMP) on expression of protein c-fos, c-jun of JNK signal transduction pathway in hippocampal neurons of anoxia/reoxygenation (A/R) rats. Methods Hippocampal neurons cultured for 7 days were randomly assigned into 6 groups (n=6):group N (no anoxia group), group C (TMP 0μg/mL), group S (10μmol/L of JNK inhibitor SP600125), group L (TMP 60μg/mL), group M (TMP 200μg/mL), and group H (TMP 800μg/mL). After incubating for 1 h, A/R model was prepared. The apoptosis rate of hippocampal neurons, and contents of protein c-fos and c-jun was detected. Results ①The apoptosis rate of hippocampal neurons decreased significantly in TMP pre-administration groups (P<0.01).②The expression of protein c-fos, c-jun decreased significantly in TMP pre-administration groups (P <0.01). Conclusions TMP has protective effect for hippocampal neurons of A/R rats through the inhibition of expression of proteins c-fos and c-jun associated with JNK signal transduction pathway.
